Application:
Simons citrate medium is used for the citrate utilization test of intestinal bacteria (GB/T4789.28-2003, 3. 5, GB/T4789.5-2003 and "People's Republic of China Pharmacopoeia" 2005 edition).
Principle:
Sodium chloride maintains a balanced osmotic pressure; magnesium ion is a cofactor in various metabolisms; ammonium dihydrogen phosphate provides a nitrogen source; dipotassium hydrogen phosphate is a buffer; sodium citrate is used as a carbon source; agar is a coagulant for the medium. Bromothymol blue is a pH indicator, which is yellow in acidity and blue in alkali.
When the bacteria can use the ammonium salt as the nitrogen source and use the citrate as the carbon source, it can grow on the citrate medium, decompose the sodium citrate, and form sodium carbonate, so that the alkali of the medium becomes blue.
Instructions:
1. Weigh 27.3g of this product, add 1 L of distilled water or deionized water, stir to dissolve and boil until completely dissolved, dispense the test tube, autoclave at 121 °C for 15 min. Take it out and place it as a slope, and wait for it to solidify.
2. Pick a small amount of fresh pure culture of the test bacteria, inoculate on the inclined surface of the medium, and incubate at 36±1°C for 1-4 days.
3. Observe the results day by day. The positive reaction was that the slant medium changed from green to blue, and the lawn was grown; aseptic growth, the slope was still green and the reaction was negative
